 Poly-lysine coating of glass bottom dishes and glass bottom plates

Materials :

 Poly-lysine solution

 Both poly-L-lysine and poly-D-lysine of MW between 30,000 - 150,000 can be used for coating.

 Poly-D-lysine is  recommended since it is not subjected to protease released from cells.

 Prepare 1mg/ml stock solution by dissolving 100mg Poly-D-lysine hydrobromide(Sigma P-0899) in 100 ml water.

 Filter  sterilize the solution then store aliquots at –20℃.

 Working solution is obtained by diluting the stock solution with sterile water to around 0.1mg/ml (0.05-0.5mg/ml).

Procedure :

 1. Add working solution of poly-lysine into a glass bottom dish or glass bottom plate.

     If cells will be cultured only on the glass surface, then only the glass surface needs to be covered with

     poly-lysine solution.

 2.Incubate for 1 hour at room temperature in a hood.

 3.Remove poly-lysine solution. Poly-lysine solution can be re-used for a few times(depending on its concentration).

 4.Rinse 3 times with PBS

 5.Poly-lysine coated glass bottom dish or glass bottom plate can be kept at 4℃ for about a month.

    Note that coating on glass surface is less stable than coating on plastic tissue culture dishes or plates.

    Thus it is recommended to coat the glass bottom dish/plate right before cell culture.

 

 Collagen coating of glass bottom dishes and glass bottom plates

Materials :

 Rat tail collagen (type I) solution

 Rat tail collagen I is available from many commercial vendors, such as BD Biosciences(cat# 354249),

 Sigma (cat #C3867), Invitrogen (cat #A10483-01).

 It is recommended to purchase collagen solutions, so that sterilization is not needed since filter sterilization

 of collagen could be problematic.

 Dilution the stock solution to 50 ug/ml with 0.02M acetic acid or 0.01M HCl.

Procedure :

 1.Add solution (50 ug/ml) into a glass bottom dish or glass bottom plate at 5 ug/cm2.

    If cells will be cultured only on the glass surface, then only the glass surface needs to be covered with collagen

    solution.

 2.Incubate for 1 hour at room temperature in a hood.

 3. Carefully aspirate the collagen solution.
     (alternatively, the glass bottom dish/plate can be left in the hood to dry overnight, and the following

     washing step is not needed)

 4. Rinse 3 times with PBS or serum-free media.

 5. Collagen coated glass bottom dish or glass bottom plate can be air dried and kept at 4℃ for about a month.

     Note that coating on glass surface is less stable than coating on plastic tissue culture dishes or plates.

     Thus it is recommended to coat the glass bottom dish/plate right before cell culture.

 

   Fibronectin coating of glass bottom dishes and glass bottom plates

Materials :

 Fibronectin solution

 Human Fibronectin from BD Biosciences(cat# 354008) is diluted to 10-50ug/ml with Ca++ and Mg++ free PBS.

 The concentration should be proportional to coating concentration, i.e, make 10ug/ml solution for 1ug/cm2

 coating, 50ug/ml for 5ug/cm2 coating, etc.

 Make appropriate aliquots and store at -20℃.

 Do not store in frost-free freezer, avoid multiple freeze thaws.

Procedure :

 1.Add appropriate amount of fibronectin solution to culture surface.

    If cells will be cultured only on the glass surface, then only the glass surface needs to be covered with the

    fibronectin solution.

 2. Incubate for 1 hour at room temperature.

 3. Carefully aspirate the remaining solution.

 4. Rinse with media or PBS.

 5. Fibronectin coated glass bottom dish or glass bottom plate should be used immediately.

 What kind of adhesive is used to adhere the dish and the cover glass? How can I detach the bottom glass?

 We use a USP class VI adhesive to adhere the dish and the cover glass.

 This adhesive is nontoxic, and it is resistant to water and common fixing reagents such as methanol and ethanol.

 This makes our glass bottom dishes and glass bottom plates ideal for long term cell culture.

 The bad news is that the adherence between the dish and the cover glass is so strong, that it is very difficult to

 detach the cover glass from the dish.

 What is the difference between the plastic surface and glass surface for growing cells?

 Most plastic vessels used to culture attached cells are made from polystyrene.

 Since polystyrene is hydrophobic and most cultured cells can not attach to hydrophobic surface, plastic cell culture vessels

 are usually surface treated to introduce hydrophilic groups onto the surface to help the attachement of cultured cells.

 Glass surface on the other hand, does not need any pre-treatment to allow the attachment of the cultured cells.

 However the attachment usually is not as strong as that of surface treated polystyrene, our experience indicates that

 poly-lysine, collagen coating works much better on plastic than on glass. Consequently, poly-lysine or collagen coating on

 glass are less stable than poly-lysine or collagen coating on surface treated plastics.
